Nanosensor Deteksi Wabah dan Infeksi

Tim peneliti pada Institute for Global Food Security (GFS), Queen’s University Belfast di Irlandia Utara, menemukan enzim uji-biomarker (enzyme biomarker test) yang murah, peka, dan cepat. Uji bio-marker enzim itu dapat melacak keberadaan penanda-enzim (enzyme markers) wabah protease – enzim pengurai protein dan peptida—pada manusia, hewan, dan produk pangan. Hasil riset itu dirilis oleh jurnal Nano Research edisi Januari 2019.

“Not only is the test cheap to produce, but it can be used anywhere and is not reliant on laboratory conditions. Eliminating the need to carry out tests in a laboratory setting is life-changing. As well as being cost-effective, it means faster diagnosis,” ungkap Dr. Claire McVey, co-author karya ilmiah itu. Hal ini juga diakui oleh Profesor Cristopher Elliott, pendiri GFS dan peneliti studi ilmiah ini : “The ability to diagnose disease or contamination quickly can have a huge impact on how serious problems can be dealt with. The ultra-low cost of the system will help reduce costs of testing and could transform the amount of testing performed in the developing world.” (Queen's University Belfast, 17/1/2019)

Tim ahli itu menciptakan metode deteksi sangat peka, cepat dan murah terhadap protease dalam susu dan urin. Metode nanosensor berbasis nanopartikel emas itu dapat menunjukkan keberadaan protease melalui reaksi perubahan warna. Nanopartikel mempercepat oksidasi kimia tetramethylbenzidine (TMB) yang terlihat melalui pembentukan warna biru-cerah. Jika protease ‘menelan’ barir casein (protein susu atau keju) pelindung, permukaan nano-artikel emas tersingkap. Jika TMB bertambah, protease mengurai casein. Sehingga oksidasi terjadi, yang memicu perubahan warna dengan cepat (Queen's University Belfast, 17/1/2019).

“When we add TMB to the casein-covered gold nanoparticles, we can tell virtually instantly if proteases are present by whether or not the solution turns blue. Normally such testing takes much longer. Using molecules other than casein to coat the surface has the potential to detect other types of enzyme biomarkers. For example, coating the nanoparticles with lipids could detect the lipase enzyme, which could help in the diagnosis of diseases such as pancreatitis,” ungkap Dr. Cuong Cao, ketua tim riset (Dr Cuong Cao, 17/1/2019).
